Проводимость монослоя кардиомиоцитов при влиянии кардиотоксичных веществ

Целью исследования явилось изучить характеристики проводимости in vitro под воздействием гептанола, этанола, эритромицина, дифенгидрамина, циклофосфамида методом оптического картирования, а также установить связь с их влиянием на потенциал-зависимые ионные каналы и щелевые контакты. Методы исследования: оптическое картирование монослоя кардиомиоцитов с флуоресцентным кальций-зависимым красителем, иммуноцитохимия, конфокальная микроскопия. Результаты: 1. Гептанол (0,1–0,9 мМ) и этанол (17–614 мМ) дозозависимо уменьшают скорость проводимости и различно действуют на величину критического радиуса кривизны фронта волны возбуждения в монослое неонатальных крысиных кардиомиоцитов. 2. Эритромицин (15–45 µМ) уменьшает скорость проводимости незначительно, максимально на 12±9%, уменьшает максимальную усваиваемую частоту максимально на 28±12% и вызывает ре-ентри на остром конце линейного препятствия в 33% случаев в монослое человеческих кардиомиоцитов, полученных из ИПСК здорового донора линии m34Sk3. Обнаружено отсутствие перечисленных эффектов эритромицина при воздействии на монослой неонатальных крысиных кардиомиоцитов. 3. Дифенгидрамин (0,3–16 µМ) уменьшает скорость проводимости максимально на 47±18%, уменьшает максимальную усваиваемую частоту максимально на 48±12% и вызывает ре-ентри на остром конце линейного препятствия в 50% случаев в монослое человеческих кардиомиоцитов, полученных из ИПСК здорового донора линии m34Sk3. 4. Циклофосфамид (213–852 µМ) не влияет на скорость проводимости в пределах погрешности, уменьшает максимальную усваиваемую частоту максимально на 25±7%, не вызывает ре-ентри на остром конце линейного препятствия в монослое человеческих кардиомиоцитов, полученных из ИПСК здорового донора линии m34Sk3. Циклофосфамид не влияет на скорость проводимости в пределах погрешности, уменьшает максимальную усваиваемую частоту максимально на 33±9% на модели неонатальных крысиных кардиомиоцитов. Обнаружено дозозависимое уменьшение площади проводимости монослоя кардиомиоцитов при влиянии (213–852 µМ) циклофосфамида и разрушение α-актинина при воздействии 213 µМ циклофосфамида на изолированные ИПСК-КМ линии m34Sk3 и НККМ до 30 минут. Новизна и применение результатов: впервые получены зависимости скорости проводимости и критического радиуса кривизны фронта волны возбуждения от концентраций гептанола и этанола и значения характеристик проводимости (скорость, максимальная усваиваемая частота, вероятность формирования ре-ентри на остром конце линейного препятствия) под влиянием эритромицина, дифенгидрамина и циклофосфамида. Монослой кардиомиоцитов, полученных из ИПСК здорового донора, приближена к реальной сердечной ткани пациента, что позволило дополнить известные данные о кардиотоксичности данных лекарственных средств и раскрыть механизм образования аритмий под их воздействием in vitro. Данному исследованию можно найти применение в медицине и разработке лекарственных средств.

Conductivity of cardiomyocyte monolayer under the influence of cardiotoxic substances

Conductivity characteristics were studied under the influence of heptanol, ethanol, erythromycin, diphenhydramine, cyclophosphamide by optical mapping in vitro, as well as a connection with their effect on voltage-gated ion channels and gap junctions. Research methods: optical mapping of a monolayer of cardiomyocytes with a fluorescent calcium-dependent dye, immunocytochemistry, confocal microscopy. Results: 1. Heptanol (0.1–0.9 mM) and ethanol (17-614 mM) dose-dependently reduce the conduction velocity and have different effects on the value of the critical radius of curvature of the excitation wave front in the monolayer of neonatal rat cardiomyocytes. 2. Erythromycin (15-45 µM) reduces the conduction velocity slightly, by a maximum of 12± 9%, reduces the maximum capture rate by a maximum of 28± 12% and causes re-entry at the sharp end of the linear obstacle in 33% of cases in a monolayer of human cardiomyocytes obtained from the IPSC of a healthy donor of the m34Sk3 line. The absence of the listed effects of erythromycin was found when exposed to a monolayer of neonatal rat cardiomyocytes. 3. Diphenhydramine (0.3–16 µM) reduces the conduction velocity by a maximum of 47±18%, reduces the maximum capture rate by a maximum of 48± 12% and causes re-entry at the sharp end of the linear obstacle in 50% of cases in a monolayer of human cardiomyocytes obtained from the IPSC of a healthy donor of the m34Sk3 line. 4. Cyclophosphamide (213-852 µM) does not affect the conduction velocity within the margin of error, reduces the maximum capture rate by a maximum of 25 ± 7%, does not cause re-entry at the sharp end of the linear obstacle in the monolayer of human cardiomyocytes obtained from the IPSC of a healthy donor of the m34Sk3 line. Cyclophosphamide does not affect the conduction velocity within the margin of error, reduces the maximum capture rate by a maximum of 33 ± 9% on a model of neonatal rat cardiomyocytes. A dose-dependent decrease in the conduction area of the cardiomyocyte monolayer was detected under the influence of (213-852 µM) cyclophosphamide and the disruption of α-actinin under the influence of 213 µM cyclophosphamide on isolated IPSC-cardiomyocites from lines m34Sk3 and neonatal rat ventricular myocytes for up to 30 minutes. Novelty and application of the results: for the first time, the dependences of the conduction velocity and the critical radius of curvature of the excitation wave front on the concentrations of heptanol and ethanol and the values of the conductivity characteristics (velocity, maximum capture rate, probability of re-entry formation at the sharp end of a linear obstacle) under the influence of erythromycin, diphenhydramine and cyclophosphamide were obtained. The monolayer of cardiomyocytes obtained from the IPSC of a healthy donor is close to the real heart tissue of the patient, which made it possible to expand the known data on the cardiotoxicity of these drugs and reveal the mechanism of arrhythmia formation under their influence in vitro. This research can be used in medicine and drug development.
