Дивергенции зарождающихся хемоавтотрофных путей фиксации со2 в гидротермальных условиях

Фиксация неорганического углерода в органический материал является химической основой функционирования первых самовоспроизводящихся С–Н–О систем в подводных гидротермальных системах на древней Земле. Там создавались благоприятные термодинамические и кинетические условия для этого процесса благодаря оптимальному составу минералов – катализаторов и термическим и химическим градиентам, возникающим в результате импульсов водородной дегазации жидкого ядра и взаимодействию, порода-вода. По современным представлениям в гидротермальных условиях архея создавался пул карбоксильных и кетокислот – автокатализаторов [напр., Braakman, Smith, 2012; Goldford et al., 2017; Маракушев, Белоногова, 2009, 2021], которые в парагенезисе с углеводородами на поверхности минералов трансформировался в автокаталитическую сеть фиксации СО2.

В представляемой работе обосновывается существование обратимых метастабильных равновесий между не метановыми углеводородами и продуктами их окисления (карбоновыми кислотами, спиртами и альдегидами) в гидротермальных условиях вулканических выбросов. Предполагается, что на древней Земле в этих условиях сформировался пул прото-метаболитов трехкомпонентной системы С-О-Н, находящихся в обратимых метастабильных равновесиях друг с другом и обладающих способностью ассимилировать СО2 из окружающей среды. Эволюция этого пула интермедиатов зарождающегося метаболизма регулировалась обратимыми шунтами - узлами бифуркации, определяющими альтернативные направления развития в сетях химических реакций и приводящие к формированию различных прото-метаболических путей хемоавтотрофной фиксации СО2. Парагенетический анализ системы С–Н–О в гидротермальном окружении, основанный на методе термодинамических потенциалов (Marakushev, Belonogova, 2009), применен для исследования центральных химических переключателей (шунтов) бифуркации электронов прото-метаболической сети определяющих химическую эволюцию пяти зарождающихся бактериальных циклов: восстановительный пентозо-фосфатный (RPP) цикл, ацетогенный и метаногенный Вуда-Лундгвала (WL) путь, восстановительный цитратный (RC) цикл, 3-гидроксипропионатный (3-HP) би-цикл. Управляемое давлением водорода равновесие фумарат (С4Н4О4) + Н2 = сукцинат(С4Н6О4) регулирует развитие прото-метаболической сети в направлении образования RC цикла или 3-HP би-цикла. Увеличение давления водорода приводит к увеличению концентрации сукцината, инициирующего образование RC цикла, а его уменьшение инициирует 3-HP би-цикл. Эволюция циклов также управлялась химическим потенциалом СО2, что отражено в работе на схеме дивергенции модулей прото-метаболических циклов. Такое зависимое от парциального давления СО2 переключение в обратную сторону направления потока электронов в RC цикле недавно было впервые продемонстрировано на примере термофильной серо-редуцирующей дельта-протеобактерии Hippea maritima [Steffens et al., 2021].

Интересная особенность бактериального автотрофного метаболизма – способность переключать СО2- фиксирующий метаболизм с WL пути на RPP цикл и наоборот была недавно открыта в термофильной анаэробной бактерии Ammonifex degensii. В зависимости от химического потенциала водорода эта бактерия организовывает свою автотрофную фиксацию углерода как WL путь или как RPP цикл [Berg et al., 2022], что регулируется другим эффективным водородным шунтом - равновесной парой пируват (С3Н4О3) +Н2 = 1,5 ацетат (С2Н4О2). Этот унаследованный современными микроорганизмами механизм переключения очевидно существовал и в прото-метаболических сетях, в которых обратимость фазового перехода через равновесие пируват ↔ ацетат создает возможность выбора развития между этими СО2-фиксирующими системами. Кроме того, химический потенциал молекулярного водорода окружающей гидротермальной среды определяет развитие сети в направлении двух кластеров фиксации СО2 – пируватный: (RC и RPP цикл) или ацетатный (ацетогенный и метаногенный WL путь и 3-HP би-цикл). Таким образом пируват (центральный хаб или метаболический репликатор), образуя парагенезис с ацетатом, создает важнейший шунт метаболического пула интермедиатов, с участием которого происходило зарождение и эволюция всех известных бактериальных автотрофных метаболических путей фиксации СО2.

Divergence of nascent chemoautotrophic CO2 fixation pathways under hydrothermal conditions

The fixation of inorganic carbon into organic material is the chemical basis for the functioning of the first self-reproducing С–Н–О systems in underwater hydrothermal systems on ancient Earth. Favorable thermodynamic and kinetic conditions for this process were created there due to the optimal composition of minerals - catalysts and thermal and chemical gradients resulting from pulses of hydrogen degassing of the liquid core and rock-water interaction. According to modern concepts, under the hydrothermal conditions of the Archean, was created a pool of carboxylic and keto acids, autocatalysts [for example, Braakman and Smith 2012; Goldford et al., 2017; Marakushev, Belonogova, 2009, 2021], which in paragenesis with hydrocarbons on the surface of minerals transformed into an autocatalytic network of CO2 fixation.

The present work provides evidence for the existence of reversible metastable equilibria between non-methane hydrocarbons and their oxidation products (carboxylic acids, alcohols, and aldehydes) under hydrothermal conditions of volcanic eruptions. It is assumed that on the ancient Earth, under these conditions, a pool of proto-metabolites of the three-component C-O-H system was formed, which are in reversible metastable equilibria with each other and have the ability to assimilate CO2 from the environment. The evolution of this pool of intermediates of nascent metabolism was regulated by reversible shunts - bifurcation nodes that determine alternative directions of development in networks of chemical reactions and lead to the formation of various proto-metabolic pathways of chemoautotrophic CO2 fixation. Paragenetic analysis of the С–Н–О system in a hydrothermal environment based on the method of thermodynamic potentials [Marakushev and Belonogova, 2009] was applied to study the central chemical switches (shunts) of the electron bifurcations of the proto-metabolic network that determine the chemical evolution of five nascent bacterial paths: the reductive pentose-phosphate (RPP) cycle, acetogenic and methanogenic Wood-Lundgwala (WL) pathway, reductive citrate (RC) cycle, 3-hydroxypropionate (3-HP) bi-cycle. Hydrogen pressure-driven equilibrium fumarate (C4H4O4) + H2 = succinate (C4H6O4) regulates the development of the proto-metabolic network towards the formation of the RC cycle or 3-HP bi-cycle. The evolution of cycles was also driven by the chemical potential of CO2, which is shown in the work on the scheme of divergence of proto-metabolic cycles modules. Such a CO2 partial pressure-dependent reversal of the direction of electron flow in the RC cycle was recently demonstrated for the first time in the case of the thermophilic sulfur-reducing delta-proteobacterium Hippea maritima as an example [Steffens et al., 2021].

An interesting feature of bacterial autotrophic metabolism, the ability to switch CO2-fixing metabolism from the WL pathway to the RPP cycle and vice versa, was recently discovered in the thermophilic anaerobic bacterium Ammonifex degensii. Depending on the chemical potential of hydrogen, this bacterium organizes its autotrophic carbon fixation as a WL pathway or as an RPP cycle [Berg et al., 2022], which is regulated by another efficient hydrogen shunt, the equilibrium pair pyruvate (С3Н4О3) + Н2 = 1.5 acetate (C2H4O2). This switching mechanism, inherited by modern microorganisms, apparently also existed in proto-metabolic networks, in which the reversibility of the phase transition through the pyruvate ↔ acetate equilibrium creates the possibility of choosing the development between these CO2-fixing systems. In addition, the chemical potential of molecular hydrogen in the surrounding hydrothermal environment determines the development of the network in the direction of two CO2 fixation clusters - pyruvate: (RC and RPP cycle) or acetate (acetogenic and methanogenic WL pathway and 3-HP bi-cycle). Thus, pyruvate (the central hub or metabolic replicator), forming a paragenesis with acetate, creates the most important shunt of the metabolic pool of intermediates, with the participation of which the origin and evolution of all known bacterial autotrophic metabolic pathways of CO2 fixation occurred.
