Влияние липидного состава липидных капель на эффективность их безбелкового слияния

Работа посвящена изучению слияния липидных капель (ЛК) — органелл, состоящих из ядра жирных кислот, таких как триолеин, окруженного монослоем фосфолипидов. Ввиду сравнительно простой реализации и универсальности воздействия липидного состава оболочек ЛК на эффективность их слияния направленное изменение липидного состава представляется удобным инструментом для перспективной терапии заболеваний, связанных с нарушением метаболизма.

В рамках работы изучалось безбелковое слияние липидных капель, приводящее к объединению их монослойных оболочек. Этот процесс требует преодоления энергетического барьера E, связанного с топологической перестройкой сливающихся липидных монослоев. Нами была произведена оценка энергетического барьера и исследовано влияние липидного состава на высоту этого барьера.

Изменение липидного состава моделировали как добавление диолеоилфосфатидилэтаноламина (ДОФЭ) к мембране, состоящей из диолеоилфосфатидилхолина (ДОФХ). Высоту E вычисляли с помощью теории упругости липидных мембран и методов молекулярной динамики. Для этого нами было сделано обобщение теории бислойного слияния [1] на случай монослойного слияния. Кроме того, высоту E определяли экспериментально по данным динамического светорассеяния (ДРС) об эволюции среднего размера частиц ЛК при различных температурах. Системы для экспериментов готовили по методике [2,3], а Е оценивали в рамках модели коагуляции, описанной в [4–6].

Сравнение теоретических и экспериментальных результатов указывает на общую тенденцию, соответствующую бислойному слиянию: увеличение доли ДОФЭ в составе мембраны приводит к понижению высоты барьера на слияние, что регистрируется как увеличение среднего размера липидных капель в эксперименте. Барьер на слияние липидных капель с оболочкой из чистого ДОФХ оказывается достаточно высоким (больше 30 kBT), чтобы обеспечить стабильность капель в системе. При этом данные молекулярной динамики свидетельствуют о том, что конечное состояние системы энергетически более устойчиво, чем в случае бислойного слияния.

Полученные результаты могут лечь в основу создания эффективных методик терапии и профилактики различных патологий, связанных с нарушением метаболизма, которые будут основаны на специфических диетах со строго определенным липидным составом липидных капель.

Influence of the lipid composition of lipid droplets on the efficiency of their protein-free fusion

The work is dedicated to the study of the fusion of lipid droplets — organelles consisting of a core of fatty acids, such as triolein, surrounded by a monolayer of phospholipids. In view of the relatively simple implementation and universality of the effect of the lipid composition of the LC membranes on the efficiency of their fusion, a targeted change in the lipid composition seems to be a convenient tool for promising therapy of diseases associated with metabolic disorders.

As part of the work, we studied the protein-free fusion of lipid droplets, leading to the unification of their monolayer shells. This process requires overcoming the energy barrier E associated with the topological rearrangement of merging lipid monolayers. We have evaluated the energy barrier and studied the effect of lipid composition on the height of this barrier.

The change in lipid composition was modeled as the addition of dioleoylphosphatidylethanolamine (DOPE) to a membrane composed of dioleoylphosphatidylcholine (DOPC). The height E was calculated using the theory of elasticity of lipid membranes and molecular dynamics methods. To this end, we generalized the theory of bilayer fusion [1] to the case of monolayer fusion. In addition, the height E was determined experimentally from dynamic light scattering (DLS) data on the evolution of the average size of lipid droplet particles at different temperatures. Systems for experiments were prepared according to the procedure [2, 3], and E was estimated within the framework of the coagulation model described in [4–6].

Comparison of theoretical and experimental results indicates a general trend corresponding to bilayer fusion: an increase in the proportion of DOPE in the membrane leads to a decrease in the height of the barrier to fusion, which is recorded as an increase in the average size of lipid droplets in the experiment. The barrier to the fusion of lipid droplets with a shell of pure DOPC is high enough (more than 30 kBT) to ensure the stability of droplets in the system. At the same time, molecular dynamics data indicate that the final state of the system is energetically more stable than in the case of bilayer fusion.

The results obtained can form the basis for the creation of effective methods for the treatment and prevention of various pathologies associated with metabolic disorders, which will be based on specific diets with a strictly defined lipid composition of lipid drops.
