Исследование механизма взаимодействия бромелина, папаина и фицина с графт-сополимерами на основе хитозана и карбоксиметилцеллюлозы

Изучение влияния микроокружения на конформационные изменения и активность ферментов является одной из фундаментальных задач современной биофизики, решение которой открывает возможности дизайна новых селективных ингибиторов или активаторов энзимов и расширяет сферы практического применения биокатализаторов. Целью настоящей работы является исследование механизма взаимодействия цистеиновых протеаз – бромелина (КФ 3.4.22.32), папаина (КФ 3.4.22.2) и фицина (КФ 3.4.22.3) – с графт-сополимерами натриевой соли карбоксиметилцеллюлозы (КМЦ) или хитозана (ХТЗ) с боковыми привитыми цепями из поли-N-винилимидазола (ПВИ) и поли-N,N-диметиламиноэтилметакрилата (ПДМАЭМА).

Конъюгаты цистеиновых протеаз с графт-сополимерами получали комплексообразованием в боратном буфере с рН 9. Механизм взаимодействия исследовали методами гибкого молекулярного докинга и ИК-спектроскопии. Содержание белка в полученных конъюгатах определяли модифицированным методом Лоури, протеолитическую активность оценивали по скорости реакции гидролиза субстрата азоказеина [1-6].

В результате исследования установлено, что со стороны графт-сополимеров во взаимодействия с молекулами цистеиновых протеаз вовлекаются как углеводный остов полисахарида, так и боковые привитые цепи и их функциональные группы. In silico исследование показало, что конъюгирование протекает самопроизвольно и характеризуется отрицательными значениями аффиности. Лиганды располагаются в каталитическом кармане глобул протеаз. Аминокислотные остатки ферментов, образующие активный центр, формируют водородные связи с графт-сополимерами на основе натриевой соли карбоксиметилцеллюлозы и хитозана с поли-N-винилимидазолом для бромелина и папаина, для фицина же наблюдается подобное взаимодействие только с графт-сополимером КМЦ-ПДМАЭМА. Кроме того, лиганды вступают в слабые физические взаимодействия, в основном – в гидрофобные, с каталитически значимыми аминокислотами. Стоит отметить, что со стороны графт-сополимеров в образовании новых связей участие принимает полисахаридный остов макромолекул графт-сополимеров, за исключением фицина, где азольные кольца привитых цепей также формируют Н-связи. Примечательно, что полярные азольные циклы, как и углеродные скелеты боковых цепей, во всех рассматриваемых случаях вступают в гидрофобные взаимодействия.

Данные ИК-спектроскопии подтверждают результаты расчетов молекулярного докинга, указывая на те же функциональные группы и фрагменты макромолекул графт-сополимеров как вовлеченные во взаимодействие с ферментами. Кроме того, отмечается смещение положения полосы Амид I в ИК-спектрах конъюгированных энзимов по сравнению с нативными. С учетом этого факта можно сделать вывод о том, что глобулы ферментов претерпевают конформационные изменения, принимая более вытянутые формы. В случаях конъюгатов с носителями на основе хитозана отчетливо выявляется уменьшение областей, содержащих α-спирали, и увеличение регионов β-складчатостей.

На основании проведенных исследований можно сделать вывод о том, что подобные структурные изменения могут оказать значительное влияние на каталитическую способность бромелина, папаина и фицина. Поэтому следующим этапом работы являлась оценка протеолитической активности конъюгированных энзимов. В случае коньюгирования энзимов с графт-сополимерами на основе натриевой соли карбоксиметилцеллюлозы, а для папаина и фицина – и с графт-сополимером хитозана с привитыми цепями из поли-N-винилимидазола, происходит увеличение как общей, так и удельной активности конъюгированных ферментов. Хотелось бы отметить, что указанный эффект (явление гиперактивации) обусловлен не высоким содержанием энзима в препаратах, а конформационными изменениями глобул и модуляцией структуры активного центра цистеиновых протеаз.

Таким образом, изучен механизм взаимодействий бромелина, папаина и фицина с графт-сополимерами натриевой соли карбоксиметилцеллюлозы или хитозана с боковыми привитыми цепями из поли-N-винилимидазола и поли-N,N-диметиламиноэтилметакрилата. Установлено, что при взаимодействии с носителями на основе карбоксиметилцеллюлозы и сополимером хитозана с N-винилимидазолом происходит увеличение протеолитической активности ферментов.

Study of the interaction mechanism of bromelain, papain and ficin with graft copolymers based on chitosan and carboxymethylcellulose

The study of the microenvironment influence on conformational changes and activity of enzymes is one of the fundamental problems of modern biophysics solving of the one opens up possibilities for the design of new selective enzyme inhibitors or activators and expands the scope of biocatalysts practical application. The aim of this work is to study the mechanism of cysteine proteases interaction – bromelain (EC 3.4.22.32), papain (EC 3.4.22.2) and ficin (EC 3.4.22.3) – with graft copolymers of sodium salt of carboxymethylcellulose (CMC) or chitosan (Cht) side grafted chains of poly(N-vinylimidazole) (PVI) and poly(N,N-dimethylaminoethyl methacrylate) (PDMAEMA).

Conjugates of cysteine proteases with graft copolymers were obtained by complexation in borate buffer with pH 9. The interaction mechanism was studied by flexible molecular docking and FTIR. The protein content in the resulting conjugates was determined by the modified Lowry method, and the proteolytic activity was assessed by the rate of the hydrolysis reaction of the azocasein substrate [1-6].

As a result of the study, both the carbohydrate backbone of the polysaccharides and the side grafted chains are involved in interactions with cysteine protease molecules. An in silico study showed that conjugation is spontaneous and characterized by negative affinity values. Ligands are located in the catalytic pocket of protease globules. The amino acid residues of the enzymes forming the active center form hydrogen bonds with graft copolymers based on the sodium salt of carboxymethyl cellulose and chitosan with poly(N-vinylimidazole) for bromelain and papain, while for ficin, a similar interaction is observed only with the CMC-PDMAEMA graft copolymer. In addition, ligands enter into weak physical interactions, mainly hydrophobic ones, with catalytically significant amino acids. It should be noted that, on the part of graft copolymers, the polysaccharide backbone of macromolecules of graft copolymers takes part in the formation of new bonds, with the exception of ficin, where the azole cycles of grafted chains also form H-bonds. It is noteworthy that the polar azole cycles, as well as the carbon skeletons of the side chains, enter into hydrophobic interactions in all cases considered.

The FTIR data confirm the results of molecular docking pointing to the same graft copolymers’ functional groups and fragments of macromolecules as being involved in the interaction with enzymes. In addition, there is a shift in the position of the Amide I band in the FTIR spectra of conjugated enzymes compared to native ones. Considering this fact, we can conclude that enzyme globules undergo conformational changes taking on more elongated shapes. In the cases of conjugates with carriers based on chitosan, a decrease in the regions containing α-helices and an increase in the regions of β-barells are clearly revealed.

Based on the studies carried out, it can be concluded that such structural changes can have a significant effect on the catalytic ability of bromelain, papain, and ficin. Therefore, the next stage of the work was the evaluation of the conjugated enzymes proteolytic activity. In the case of conjugation of enzymes with graft copolymers based on the sodium salt of carboxymethyl cellulose, and for papain and ficin with a graft copolymer of chitosan with grafted chains of poly(N-vinylimidazole), there is an increase in both total and specific activity of conjugated enzymes. We would like to note that this effect (the phenomenon of hyperactivation) is due not to the high content of the enzyme in the preparations, but to conformational changes in globules and modulation of the structure of the active center of cysteine proteases.

Thus, the mechanism of interactions of bromelain, papain, and ficin with graft copolymers of the sodium salt of carboxymethyl cellulose or chitosan with grafted side chains of poly(N-vinylimidazole) and poly(N,N-dimethylaminoethyl methacrylate) has been studied. It has been established that the interaction with carriers based on carboxymethyl cellulose and a copolymer of chitosan with N-vinylimidazole leads to an increase in the proteolytic activity of enzymes.
