Основа метаболизма – транспорт орто-пара спиновых изомеров Н2О по аквапориновым каналам сквозь мембраны

Известно, что метаболизм, как процесс, отражает транспорт веществ в организме, главным из которых является вода из-за большой массовой доли: мозг, сердце, легкие ~80%, кровь ~90%. Физически ясно, что наиболее критическим местом транспорта воды в организме являются мембраны с аквапориновыми каналами, которые пропускают только мономеры H2O со скоростью 3Е9 молекул/с. Напомним здесь, что мономеры H2O отличаются ориентацией спина протонов и являются орто- и пара-изомерами. При этом орто-H2O имеют магнитный момент (спины параллельны) и всегда вращаются. Напротив, пара-H2O - не магнит (спины антипараллельны) и часть из них не вращается в соответствии с распределением Больцмана по вращательным состояниям. Открытие аквапориновых каналов (Peter Agre) было отмечено 20 лет назад нобелевской премией [1]. Заметим, что аквапориновый канал имеет диаметр ~0.3 нм при размере мономера H2O ~0.28 нм и дипольный ключ в середине (Рис.1). Несмотря на эти факторы, каналы мембраны почки человека могут пропустить до 200 литров воды за сутки [1]. Однако P. Agre et al. [2] отмечает, что остаѐтся неясным механизм образования цепочки из мономеров H2O внутри канала с разрывом водородных связей в окрестности дипольного ключа (Рис.1, нижняя панель). Совокупность данных, полученных за две декады в разных научных центрах, дают нам обоснование наличия спиновых изомеров и их конверсию, а также место их локализации в воде и водных растворах

50 лет назад пионерская работа Ахманова С.А. и др. [3] открыла новую эру нелинейно-оптической активной спектроскопии комбинационного рассеяния (CARS). Здесь нелинейная оптика обеспечила с помощью пучков лазеров видимого диапазона уникальную возможность изучения [4,5] движения молекул в воде и водных растворах в области гигантского поглощения на ГГц-ТГц частотах. Так методом CARS были открыты свободные вращения орто-пара спиновых изомеров молекул Н2О в воде [6] и селективное связывание пара-изомеров Н2О при формировании гидратных оболочек в водных растворах белков [6]. Позднее [7] спиновые изомеры Н2О были обнаружены в воде также. Оставалось неясным, где мономеры Н2О могут быть локализованы в воде и гидратных оболочках?

Ранее [8] мы обосновали, что льдоподобные структуры льда Ih в воде [9-11] и гидратных оболочках [12-15] способны локализовать мономеры Н2О, как в фуллерене [16], в гексагональных полостях вдоль оси с, поперечный размер которых 0.57 нм почти в два раза больше диаметра аквапоринового канала [1,2]. Так было установлено [13], что упругость и прочность структуры гидратных оболочек гемоглобина внутри эритроцитов весьма велика, чтобы их разрушить давлением оболочки. Только расплав структуры [13] из-за пара-орто конверсии Н2О [12] сопровождается потерей ~55% воды через аквапориновые каналы и обеспечивает деформацию эритроцитов для перемещении по капиллярам. Подобный расплав (пара-орто конверсия Н2О) гидратных оболочек (до 90% воды) белка лизоцима имеет место в курином яйце в инкубаторе при температуре ~37.5 0С и транспорт мономеров Н2О через аквапориновые каналы мембраны желтка для орошения

ядра и последующего его деления [15], который находится наверху желтка под мембраной. Мы считаем, что ключевым фактором в таком представлении являются тепловые флуктуации, которые создают смешанные квантовые состояния близко расположенных вращательных уровней и обеспечивают пара-орто спиновую конверсию. Затем вращающиеся орто-пара спиновые изомеры Н2О проходят сквозь дипольный ключ аквапоринового канала мембраны [1,2] со скоростью 3Е9 молекул/с без остановки и поддерживают метаболизм организма на необходимом уровне.

The basis of metabolism is the transport of ortho-para spin isomer-H2O via aquaporin channels across membranes

It is known that metabolism, as a process, reflects the transport of substances in the body, the main of which is water due to the large mass fraction: brain, heart, lungs ~ 80%, blood ~ 90%. It is physically clear that the most critical site of water transport in the body are membranes with aquaporin channels, which allow only H2O monomers to pass through at a rate of 3E9 molecules/s. We recall here that the H2O monomers differ in the orientation of the proton spin and are ortho- and para-isomers. At the same time, ortho-H2O have a magnetic moment (the spins are parallel) and always rotate. In contrast, para-H2O is not a magnet (the spins are antiparallel) and some of them do not rotate in accordance with the Boltzmann distribution over rotational states. The discovery of aquaporin channels (Peter Agre) was awarded the Nobel Prize 20 years ago [1]. Note that the aquaporin channel has a diameter of ~0.3 nm with a H2O monomer size of ~0.28 nm and a dipole key in the middle (Fig. 1). Despite these factors, human kidney membrane channels can pass up to 200 liters of water per day [1]. P. Agre et al. [2] notes that the mechanism of formation of a chain of H2O monomers inside the channel with the breaking of hydrogen bonds in the vicinity of the dipole key remains unclear (Fig. 1, bottom panel). The totality of data obtained over two decades in different scientific centers gives us a rationale for the presence of spin isomers and their conversion, as well as the place of their localization in water and aqueous solutions.

50 years ago, the pioneering work of Akhmanov S.A. et al. [3] opened a new era of nonlinear optical active Raman spectroscopy (CARS). Here, using laser beams in the visible range, nonlinear optics provided a unique opportunity to study [4, 5] the motion of molecules in water and aqueous solutions in the region of giant absorption at GHz–THz frequencies. Thus, free rotations of ortho-para spin isomers of H2O molecules in water [6] and selective binding of para-isomers of H2O during the formation of hydration shells in aqueous solutions of proteins were discovered by the CARS method [6]. Later [7], the spin isomers of H2O were also found in water. It remained unclear where the H2O monomers can be localized in water and hydration shells?

Early [8], we substantiated that the ice-like structures of ice Ih in water [9–11] and hydration shells [12–15] are capable of localizing H2O monomers, as in fullerene [16], in hexagonal cavities along the c axis, the transverse size of which is 0.57 nm is almost twice the diameter of the aquaporin channel [1, 2]. So it was established [13] that the elasticity and strength of the structure of hemoglobin hydration shells inside erythrocytes is very high in order to destroy them by shell pressure. Only the melt of the structure [13] due to para-ortho conversion of H2O [12] is accompanied by the loss of ~55% of water through aquaporin channels and provides deformation of erythrocytes for movement through capillaries. A similar melt (para-ortho conversion of H2O) of hydration shells (up to 90% water) of lysozyme protein takes place in a chicken egg in an incubator at a temperature of ~37.5 0C and the transport of H2O monomers through the aquaporin

channels of the yolk membrane for irrigation of the nucleus and its subsequent division [15], which is located at the top of the yolk under the membrane. We believe that the key factor in this representation is thermal fluctuations, which create mixed quantum states of closely spaced rotational levels and provide para-ortho spin conversion. Then, the rotating ortho-para H2O spin isomers pass through the dipole key of the aquaporin channel of the membrane [1, 2] at a rate of 3E9 molecules/s without stopping and maintain the body's metabolism at the required level.
